Antimelanoma activity of Heterocapsa triquetra pigments ArchiMer
Haguet, Quentin; Bonnet, Antoine; Berard, Jean-baptiste; Goldberg, Joelle; Joguet, Nicolas; Fleury, Audrey; Thiery, Valerie; Picot, Laurent.
Heterocapsa triquetra (Ht), a non-toxic dinophyte, was selected to investigate the presence of pigments inhibiting melanoma cells growth. We developed an innovative microwave-assisted extraction process (MAE), determined the Ht pigment composition using Reverse Phase-High Performance Liquid Chromatography (HPLC) and high-resolution Ultra Performance Liquid Chromatography-Mass Spectrometry E (UPLC-MSE) analysis and assessed their antiproliferative activity in human invasive melanoma cells (A2058) grown in vitro. MAE allowed a fast and efficient extraction of Ht pigments as compared with conventional processes (soaking, sonication). Seventeen pigments and derivatives were unequivocally identified by UPLC-MSᴱ in the Ht extract, among which two were supposed...

Effect of pH on the copigmentation of anthocyanins from Cabernet Sauvignon grape extracts with organic acids Scientia Agricola
Gauche,Cony; Malagoli,Elisa da Silva; Bordignon Luiz,Marilde Terezinha.
Anthocyanins show low-stability when exposed to different food processing conditions. Copigmentation is one of the main reactions contributing to the in vivo color responsible to the stability of anthocyanins. In the aim of holding the red color, copigmentation effect of organic acids (caffeic, ferulic, gallic and tannic acids) combined with anthocyanins in crude Cabernet Sauvignon (Vitis vinifera L.) grape skin extract at pH values (1.0, 2.0, 3.0, 3.3, 3.5, 3.7, 4.0, 4.5) was evaluated in this research. The maximum copigmentation effect, revealed by the hyperchromic and bathochromic shifts in anthocyanin maximum absorbance wavelength, was obtained at pH 3.3 with every acid used. Anthocyanin stability was followed by measuring the loss of color, thus it...

Heavy metal tolerance (Cr, Ag and Hg) in bacteria isolated from sewage BJM
Lima e Silva,Agostinho A. de; Carvalho,Márcia A. Ribeiro de; Souza,Sérgio A. L de; Dias,Patrícia M. Teixeira; Silva Filho,Renato G. da; Saramago,Carmen S. de Meirelles; Bento,Cleonice A. de Melo; Hofer,Ernesto.
Samples of sewage from a university hospital and a chemistry technical school were analysed for the percentage of bacterial tolerance to chromium (Cr), silver (Ag) and mercury (Hg). Additionally, we investigated the effect of these metals on pigmentation and on some enzymatic activities of the metal tolerant strains isolated, as well as antimicrobial resistance in some metal tolerant Enterobacteriaceae strains. Tolerance to Cr was observed mainly in Gram positive bacteria while in the case of Ag and Hg the tolerant bacteria were predominately Gram negative. Hg was the metal for which the percentage of tolerance was significantly higher, especially in samples from the hospital sewage (4.1%). Mercury also had the most discernible effect on color of the...

Histological characterization of cellular types during Scinax fuscovarius oogenesis (Lutz) (Anura, Hylidae) Zoologia
Oliveira,Classius de; Santos,Lia Raquel de Souza.
This paper describes morphological characteristics of the ovarian germinative cells of the Scinax fuscovarius (Lutz, 1925). The ovary is organized in primordial germinative cells (oogonia) and follicular structures (ovarian follicle) - oocytes surrounded by follicular cells. Oogonia: their nests are peripherically localized, containing cells with large and oval nucleus. Oocytes I: basophilic cytoplasm; the spherical nucleus presents few nucleoli, one or two; the follicular cells (one tenuous layer) surround these previtellogenic cells. Oocytes II: the cell is larger and the cytoplasm becomes more basophilic; the nucleus presents few nucleoli and contains many chromosomes in the periphery (beginning the perinucleolar stage). Oocytes III: the cytoplasm...

HISTOLOGICAL DESCRIPTION OF THE REPRODUCTIVE SYSTEM OF MALE AND FEMALE HATCHLINGS OF THE MAGDALENA RIVER TURTLE (Podocnemis lewyana) Acta biol.Colomb.
SÁNCHEZ-OSPINA,ANDRÉS C; RODRIGUEZ,BERARDO; CEBALLOS,CLAUDIA P..
In this study we described the macroscopic and microscopic histology of the reproductive system of male and female Podocnemis lewyana neonates (3.5 months old). We found macroscopic differences in the morphology of the gonads between the sexes, with ovaries being twice as long as testes, and testes being twice as wide as ovaries. Microscopically, we identified several immature elements, such as the lack of a muscle layer in the oviduct of females, and simple epithelia instead of pseudostratified epithelia in the oviduct and epididymis described in reptile adults. We also found a black pigment observed macroscopically in the mesovarium, and macroscopically and histologically in the epididymis. This pigment is consistent with the center of melano-macrophages...

Les facteurs de verdissement de l'huître en claires : le milieu hydrobiologique et benthique et ses variations ArchiMer
Moreau, Jean.
In a preliminary work (MOREAU, 1967), the importance of the role of pigments in fattening ponds and the part they played in the process of Navicula ostrearia B.'s pigmentation had been studied in fattening ponds situated at two different points of the Marennes-Oléron basin. The first of these fattening ponds were located in the Seudre Valley, at La Tremblade and at Eguillate, the second ones in the Daire area more widely fed by coastal waters and where the greening conditions of the fattening ponds are different from those observed in Seudre (figure 1 and table 1). 370 Dosing the potency of various chlorophylls had highlighted a noticeable constancy of chlorophyll B and, a variance for those of type A and C (table 2) which had made it possible to conclude...

Mantle histology, histochemistry and ultrastructure of the pearl oyster Pinctada margaritifera (L.) ArchiMer
Jabbour-zahab, Roula; Chagot, Dominique; Blanc, François; Grizel, Henri.
The edge and isthmus of the mantle of Pinctada margaritifera (Mollusc, Bivalvia) were studied using light and electron microscopy from a morphological and histochemical point of view. Six areas with several subareas were differentiated in the mantle edge. One of these areas is similar to the epithelium isthmus, Trials to elucidate the nature of the pigment responsible for the black colour of the shell and pearl indicate a melanin-like material. Possible functions of the distinctive epithelial areas in Pinctada margartifera mantle are discussed according to available data on the secretion of organic matrix of the shell, protein sclerotization and deposition of crystals in some pearl-forming molluscs. Three different epithelial areas in the outer mantle edge...

Molecular Pathways and Pigments Underlying the Colors of the Pearl Oyster Pinctada margaritifera var. cumingii (Linnaeus 1758) ArchiMer
Stenger, Pierre-louis; Ky, Chin-long; Reisser, Celine; Duboisset, Julien; Dicko, Hamadou; Durand, Patrick; Quintric, Laure; Planes, Serge; Vidal-dupiol, Jeremie.
The shell color of the Mollusca has attracted naturalists and collectors for hundreds of years, while the molecular pathways regulating pigment production and the pigments themselves remain poorly described. In this study, our aim was to identify the main pigments and their molecular pathways in the pearl oyster Pinctada margaritifera—the species displaying the broadest range of colors. Three inner shell colors were investigated—red, yellow, and green. To maximize phenotypic homogeneity, a controlled population approach combined with common garden conditioning was used. Comparative analysis of transcriptomes (RNA-seq) of P. margaritifera with different shell colors revealed the central role of the heme pathway, which is involved in the production of red...

Monthly HPLC measurements of pigment concentration from an intertidal muddy sediment of Marennes-Oléron Bay, France ArchiMer
Cariou-le Gall, Valerie; Blanchard, Gerard.
Chloropigments and carotenoids were measured by HPLC in an intertidal muddy sediment of Marennes-Oleron Bay (France). Concentrations were determined as a function of sediment depth at low tide. The analyses were carried out at monthly intervals over 1 yr. Pigment analysis indicated that the microphytobenthic community was dominated by diatoms throughout the sampling year. Chlorophyll b was not encountered at any time or depth, indicating that no input from macrophytic detritus had occurred at the sampling site. There was a large pool of phaeopigments a, of which phaeophorbides were the major forms (75%). A microphytobenthic bloom occurred between March and June during which phaeopigments significantly increased with a high proportion of phaeophorbides...

Morphology, ultrastructure and molecular phylogeny of Wangodinium sinense gen. et sp. nov. (Gymnodiniales, Dinophyceae) and revisiting of Gymnodinium dorsalisulcum and Gymnodinium impudicum ArchiMer
Luo, Zhaohe; Hu, Zhangxi; Tang, Yingzhong; Mertens, Kenneth; Leaw, Chui Pin; Lim, Po Teen; Teng, Sing Tung; Wang, Lei; Gu, Haifeng.
The genus Gymnodinium includes many morphologically similar species, but molecular phylogenies show that it is polyphyletic. Eight strains of Gymnodinium impudicum, Gymnodinium dorsalisulcum and a novel Gymnodinium‐like species from Chinese and Malaysian waters and the Mediterranean Sea were established. All of these strains were examined with LM, SEM and TEM. SSU, LSU and internal transcribed spacers rDNA sequences were obtained. A new genus, Wangodinium, was erected to incorporate strains with a loop‐shaped apical structure complex (ASC) comprising two rows of amphiesmal vesicles, here referred to as a new type of ASC. The chloroplasts of Wangodinium sinense are enveloped by two membranes. Pigment analysis shows that peridinin is the main accessory...

Photoacclimation in three species of freshwater red algae Braz. J. Plant Physiol.
Bautista,Anna I.N.; Necchi Jr.,Orlando.
Three freshwater Rhodophyta species (Audouinella eugenea, A. hermannii and Compsopogon coeruleus) were tested as to their responses (photosynthesis, growth and pigment concentration) to two irradiances (low light, LL, 65 µmol m-2 s-1 and high light, HL, 300 µmol m-2 s-1) and two periods (short time, ST, 4 d, and long time, LT, 28 d). Higher growth rates were consistently observed at LL but significant differences were observed only for A. hermannii. Higher values of photoinhibition at LL were found for the three species, which is consistent with the dynamic photoinhibition as a reversible photoprotective mechanism against high irradiance. Light-induced decreases of effective quantum yield (EQY) were observed in the three species consisting of pronounced...

Pigment dynamics associated with the grazing of a ciliate and a flagellate feeding on a cyanobacterium ArchiMer
Mostajir, B; Bustillos-guzman, J; Claustre, H; Rassoulzadegan, F.
The egestion of particulate material as well as pigment degradation during microzooplankton grazing on phytoplankton are poorly known processes. In an attempt to evaluate these processes, changes in pigment concentrations within various size fractions were monitored in batch cultures of an assemblage of a pelagic ciliate (Strombidium sulcatum) and a heterotrophic flagellate (Paraphysomonas sp.) feeding on a cyanobacterium (Synechococcus sp.) over a 10-day period. Chlorophyll a, carotenoids and phaeopigments were not found in the 0.1-0.7 mu m fraction while the pigments originally in the 0.7-3.0 mu m fraction (prey) were transferred into the > 3.0 mu m size fraction (predator). During this transfer, the carotenoids (zeaxanthin and B-carotene) were not...

Species-specific differences in diatom-induced anomalies in calanoid copepod embryogenesis: are they linked to the stock-piled antioxidants? IBSS Repository
Khanaychenko, A. N..
Egg production and viability of co-occurring in nature copepods, Calanus helgolandicus and Calanoides carinatus, fed diatom, Thalassiosira rotula, and then switched to dinoflagellate Prorocentrum minimum, were compared in experimental conditions. Egg production of both species on diatom diet was similar (up to 17-22 eggs.female-1.d-1) but no viable nauplii were observed. Development was arrested at different stages depending on degree of anomalies. Abnormal embryos displayed various degrees of deteriorations in pigment distribution and organization of extracellular matrix. Reproductive responses of C.carinatus both to negative (diatom) and positive (dinoflagellate) diets were postponed in comparison with quick responses of C.helgolandicus. After 3 days of...

Study of phycocyanin production from Spirulina platensis under different light spectra BABT
Walter,Alfredo; Carvalho,Júlio Cesar de; Soccol,Vanete Thomaz; Faria,Ana Bárbara Bisinella de; Ghiggi,Vanessa; Soccol,Carlos Ricardo.
The aim of this work was to investigate the production of phycocyanin by Spirulina platensis under different spectra of light. The dependent variables evaluated were the amount of phycocyanin obtained and its purity, demonstrating that there might be a restructuring of phycobilisomes, especially when the culture was subjected to red light, which increased the purity level up to 33% with a reduction of 16% in phycocyanin content, but with higher photosynthetic efficiency compared to natural light.

The influence of hydrodynamics on the spatial and temporal variation of phytoplankton pigments in a large, sub-tropical coastal lake (Brazil) BABT
Cardoso,Luciana de Souza; Marques,David da Motta.
This study evaluated the spatial and temporal distribution of phytoplankton pigments in Itapeva Lake and its relationship with hydrodynamic aspects. Regarding spatial distribution, a decreasing N®S gradient was generally observed for the pigments, except in summer. This inversion observed during the summer was influenced by the predominant fetch (N-E). The horizontal heterogeneity was proved (ANOVA) for all seasons of the year, except spring. Spatially in spring, the vertical variance was much more significant (p<0.05) than the horizontal one. The sampling shifts presented a permanent degree of variability among the seasons of the year, showing the existence of a diurnal cycle in the concentration of chlorophyll a. This behavior was related to the...

The Potential of Microalgae for the Production of Bioactive Molecules of Pharmaceutical Interest ArchiMer
Mimouni, Virginie; Ulmann, Lionel; Pasquet, Virginie; Mathieu, Marie; Picot, Laurent; Bougaran, Gael; Cadoret, Jean-paul; Morant-manceau, Annick; Schoefs, Benoit.
Through the photosynthetic activity, microalgae process more than 25% of annual inorganic carbon dissolved in oceans into carbohydrates that ultimately, serve to feed the other levels of the trophic networks. Besides, microalgae synthesize bioactive molecules such as pigments and lipids that exhibit health properties. In addition, abiotic stresses, such as high irradiance, nutrient starvation, UV irradiation, trigger metabolic reorientations ending with the production of other bioactive compounds such as omega-3 fatty acids or carotenoids. Traditionally, these compounds are acquired through the dietary alimentation. The increasing, and often unsatisfied, demand for compounds from natural sources, combined with the decrease of the halieutic resources,...
